Biochemical

Assays

Sandwich or direct ELISA:

A solid phase heterogeneous assay to determine antibody-antigen interaction. In addition, homogenous

state-of-the-art methodologies such as TRF, HTRF and Alpha Screen can be employed to increase sensitivity.

 

Competition ELISA:

Analysis of free cytotoxic molecule released from the ADC.

 

SDS-PAGE:

A traditional assay for characterization of specicity, MW and purity of antibodies.

 

Western-Blot:

Immuno-Specic identication, MW and purity of

antibodies/drugs.

 

Differential Scanning Fluorimetry (DSF):

Measures the thermal stability of a target protein by performing thermal denaturation curve in the presence of a fluorescent dye during functional characterization (Nature Protocol, 2007).

 

UV/VIS:

Drug-to-antibody ratio measurements using UV/VIS spectroscopic analysis.

 

ELISA:

Cell-based assay to determine antibody-antigen interaction.

 

Internalization:

Quantitative and qualitative analysis of antibody internalization into cells using fluorescent methods.

 

Cytotoxicity:

Cell-based assays to determine the cytotoxic eect of the ADC using colorimetric, fluorometric and luminesentic assays.

 

Tissue Distribution:

Immunohistochemical proling of antibodies using commercial available and customized Tissue Multi Arrays.
